Useful Treatment of Severe Atopic Dermatitis with Ganoderma lucidum ( reishi ) : A Multiple-Case study

Atopic dermatitis (ADE) is one of the most common chronic inflammatory skin diseases. Treatment options include the use of lubricants (emollients), antihistaminics and corticosteroids in either topical or oral forms. However, severe ADE patients are frequently recalcitrant to these medications. In this study, we investigated five out-patients suffering from severe ADE (age range: 11 -41 years) who were treated with a mushroom called reishi (Ganoderma lucidum) at a dose of 4-6 tablets daily in divided doses at a hospital. Reishi tablets containing reishi extract equivalent to 0.94 g of dried mushroom each. In addition to physical signs and observations, the severity scores, serum immunoglobulin E (IgE) and thymus and activated-regulated chemokine (TARC) levels were monitored at 3 months after reishi administration. Four of 5 patients established marked alleviation of symptoms with improved clinical indexes after 1 .5–3 months of treatment: viz., itching, flare and swelling on the face/skin were less pronounced, and excoriations and lichenification on the trunk and extremities were markedly improved after treatment. Based on our findings, reishi may serve as a valuable option in the treatment of severe ADE patients, albeit the efficacy of reishi may depend on the severity of pruritus.

Ganoderma lucidum suppresses growth of breast cancer cells through the inhibition of Akt/NF-kappaB signaling.

 Abstract

Ganoderma lucidum (Reishi, Lingzhi) is a popular Asian mushroom that has been used for more than 2 millennia for the general promotion of health and was therefore called the "Mushroom of Immortality." Ganoderma lucidum was also used in traditional Chinese medicine to prevent or treat a variety of diseases, including cancer. We previously demonstrated that Ganoderma lucidum suppresses the invasive behavior of breast cancer cells by inhibiting the transcription factor NF-kappaB. However, the molecular mechanisms responsible for the inhibitory effects of Ganoderma lucidum on the growth of highly invasive and metastatic breast cancer cells has not been fully elucidated. Here, we show that Ganoderma lucidum inhibits proliferation of breast cancer MDA-MB-231 cells by downregulating Akt/NF-kappaB signaling. Ganoderma lucidum suppresses phosphorylation of Akt on Ser473 and downregulates the expression of Akt, which results in the inhibition of NF-kappaB activity in MDA-MB-231 cells. The biological effect of Ganoderma lucidum was demonstrated by cell cycle arrest at G0/G1, which was the result of the downregulation of expression of NF-kappaB-regulated cyclin D1, followed by the inhibition of cdk4. Our results suggest that Ganoderma lucidum inhibits the growth of MDA-MB-231 breast cancer cells by modulating Akt/NF-kappaB signaling and could have potential therapeutic use for the treatment of breast cancer.

نتایج ما نشان می دهد که Ganoderma lucidum با تعدیل سیگنالینگ Akt / NF-kappaB مانع رشد MDA-MB-231 سلولهای سرطانی پستان می شود و می تواند استفاده درمانی بالقوه برای معالجه سرطان پستان داشته باشد.

Abstract

In this study, we investigated the effects of the aqueous extracts of Lingzhi or Reishi medicinal mushroom, Ganoderma lucidum, obtained from three localities (China; and Morelos and Michoacan, Mexico) on cervical cells transformed by human papillomavirus (HeLa and SiHa) and C-33A cancer cells. The cells were plated in DMEM medium supplemented, and were incubated in the presence of different concentrations of G. lucidum for 24 h. Cell proliferation was determined by MTT colorimetric assay and viability by trypan blue assay. Inhibitory dose was determined (IC50) of the three different extracts of G. lucidum in the culture cell lines mentioned above. The apoptosis process was confirmed by nuclear DNA fragmentation and the cell cycle was determined by flow cytometry. The results showed that aqueous extracts G. lucidum obtained from three localities produced inhibition in the proliferation of VPH transformed cells; they also induced apoptosis and cell cycle arrest in HeLa, SiHa, and C-33A cancer cells. Therefore, it was found that aqueous extracts G. lucidum obtained from three different locations produced inhibitory effect on cancer cells and may have a potential therapeutic use for the prevention and treatment of this disease.

Control of oral human papillomavirus (HPV) by medicinal mushrooms, Trametes versicolor and Ganoderma lucidum: a preliminary clinical trial.

Abstract

This preliminary randomized study investigated the efficacy of medicinal mushrooms, Trametes versicolor (TV), Ganoderma lucidum (GL), and Laetiporus sulphureus (LS), on the clearance of oral human papillomavirus (HPV, serotypes 16 and 18). Among 472 patients who underwent oral swabs for gingivitis, 61 patients were positive for HPV16 or HPV18. Twenty patients were included in group 1 (LS) and 41 patients were included in group 2 (TV+GL) for 2 months. Polymerase chain reaction (PCR) for HPV was performed at inclusion and after 2 months. In group 1, the clearance was equal to 5% after 2 months of treatment. In group 2, the clearance was equal to 88% (P<0.001). The detection of HPV16 or HPV18 could become relevant in routine since positivity is frequent and because a harmless and costless treatment may exist. The use of TV+GL for the clearance of oral HPV deserves further investigation.

نتایج این تجقیق نشان داد که استفاده از قارچ های استفاده شده به میزان ۸۸ درصد سبب منفی شدن ویروس در افراد مورد مطالعه شد.
