مقاله 2013: بهینه سازی شرایط کشت استرین های وحشی اروپایی قارچ دکمه ای دارویی و کولتیوار های برزیلی Optimization of the cultivation conditions for mushroom production with European wild strains of Agaricus subrufescens and Brazilian cultivarsAbstractBACKGROUNDThe almond mushroom Agaricus subrufescens (formerly Agaricus blazei or Agaricus brasiliensis)
is cultivated at commercial level in Brazil and some Asian countries on
local substrates and casing mixtures. Despite its tropical origin, A. subrufescens
might be a seasonal option for mushroom growers in western countries,
where some wild strains have been isolated. For this purpose,
cultivation conditions were developed starting from the substrate and
casing mixture commonly used for commercial production of the button
mushroom Agaricus bisporus in France. RESULTSThe commercial compost, based on wheat straw and horse manure, used for A. bisporus and the casing mixture (peat and limestone) supplemented with fine sand proved efficient to grow A. subrufescens.
Increasing the depth of the casing layer improved significantly the
yield and time to fruiting. Daily variations in temperature did not
markedly modify the yield. Significantly higher mushroom biomass was
obtained with three wild European strains compared with three Brazilian
cultivars. The very productive wild strain CA438-A gave mushrooms of
size and dry matter content comparable to those of a cultivar. CONCLUSIONCommercial production of A. subrufescens can be developed in western countries on the wheat straw-based substrate commonly used for A. bisporus
in these regions, by a simple modification of the casing mixture and
maintaining the incubation temperature throughout the crop, which is
expected to save energy during summer. Good yields were obtained
cultivating European strains under optimised parameters. © 2013 Society

Maitake (Grifola frondosa)
was hydrothermally extracted at 4 different temperatures (121, 130,
140, and 150°C) for 30 and 60 min, and the total phenolic content (TPC),
antioxidant activity, and β-glucan content of the extracts was
evaluated. The highest TPC was detected in the extract at 150°C treated
for 60 min with 13.61 mg gallic acid equivalents (GAE)/g. The
antioxidant activity of the maitake extracts, evaluated by determining
DPPH radical scavenging activity (RSA), ABTS RSA, reducing power, and
tyrosinase inhibitory activity increased with increasing treatment
temperature and time. The content of β-glucan was the highest (5.13%) in
the extract prepared at 140°C for 30min. These results suggest that
hydrothermal extraction could be used as a tool to increase the
antioxidant activity of maitake extracts.

Drying effects on the antioxidant properties of polysaccharides obtained from Agaricus blazei MurrillAbstractThree polysaccharides (ABMP-F, ABMP-V, ABMP-A) were obtained from Agaricus blazei Murrill
via methods such as freeze drying, vacuum drying and air drying,
respectively. Their chemical compositions were examined, and antioxidant
activities were investigated on the basis of assay for hydroxyl
radical, DPPH radical, ABTS free radical scavenging ability and assay
for Fe2+_chelating ability. Results showed that the three
ABMPs have different physicochemical and antioxidant properties.
Compared with air drying and vacuum drying methods, freeze drying method
resulted to ABMP with higher neutral sugar, polysaccharide yield,
uronic acid content, and stronger antioxidant abilities of hydroxyl
radical, DPPH radical, ABTS radical scavenging and Fe2+-chelating. As a result, Agaricus blazei Murrill
polysaccharides are natural antioxidant and freeze drying method serves
as a good choice for the preparation of such polysaccharides and should

have been studied for precisely characterizing it and defining its
putative involvement in thermo-tolerance of both vegetative mycelium
growth and sporophore differentiation. Sequence polymorphism was
analyzed in 3 homokaryons of A. bisporus and 24 strains having
different abilities for mycelial growth at temperatures above 30 °C and
for producing mature fruiting bodies at 25 °C. The level of gene
expression was measured by real-time PCR both in vegetative mycelium
after transfer from 25 to 32 °C and in primordia and fruiting bodies
produced during cultures at 17 or 25 °C. The results indicated that aap1 gene belong to a new subfamily of the yeast YAP1 homologs. It is not a dominant contributor to the thermo-tolerance of A. bisporus,
but the protein it encodes may be involved as an overall stress
resistance transcription factor. The way Aap1 senses redox level differs
from that of AP-1-like transcription factor Yap1.

utilization. Agricultural and non-agricultural disposal of the spent
mushroom substrate, is possible when it is secured in a suitable manner,
minimizes the risk of transmission of pests, fungi, bacteria, vinises,
and weed seeds. After application into the soil SMS improves its
physical properties and increases the content of macro- and
microelements.

Abstract This study
assessed the application of an antibrowning solution using vacuum
impregnation (VI) and then electron-beam irradiation as a means to
extend the shelf life of sliced white button mushrooms (Agaricus bisporus).
A preliminary study helped to determine the best antibrowning solution
and VI process parameters. Mushroom slices were impregnated with 2 g/100
g ascorbic acid + 1 g/100 g calcium lactate; 2 g/100 g citric acid + 1
g/100 g calcium lactate; 1 g/100 g chitosan + 1 g/100 g calcium lactate;
and 1 g/100 g calcium lactate at different vacuum pressures and times
and atmospheric restoration times. Selection of the antibrowning
solution and VI parameters was based on texture and color of the
mushroom slices. Next, the slices were irradiated at 1 kGy using a
1.35-MeV e-beam accelerator. Physicochemical, sensory, and microbial
quality of mushrooms was monitored for 15 d at 4 °C. The best
impregnation process in this study was 2 g/100 g ascorbic acid and 1
g/100 g calcium lactate at 50 mm Hg for 5 min and an atmospheric
restoration time of 5 min. The control (untreated) samples suffered
structural losses throughout storage. Only the vacuum
impregnated-irradiated samples had acceptable color by the end of
storage. Sensory panelists consistently preferred the samples produced
with VI and irradiation because exposure to ionizing radiation inhibited
growth of spoilage microorganisms. Practical Application Extending
the shelf life of mushrooms is important for their marketing and
distribution and reliable preservation methods are still needed.
Treating sliced mushrooms with vacuum impregnation and electron-beam
irradiation introduces physiologically active components such as
beneficial antioxidants, vitamins, and cations, while assuring safety
and maintaining quality.

Pleurotus florida [P. ostreatus] was grown on
different substrates, i.e. paddy straw, sunflower head, sunflower stalk
and jackfruit rind. The weight of the different substrates was recorded
at 7, 14 and 21 days after inoculation. The highest weight loss was
observed in sunflower stalk at 7 days (1.73 g), 14 days (2.51 g) and 21
days (3.87 g) after spawning. The lowest weight loss was observed on
jackfruit rind at 7 days (1.16 g), 14 days (1.91 g) and 21 days (2.65 g)
after spawning. The highest yield (354 g/bag) and bio-efficiency (53.79
%) of P. florida was recorded in sunflower stalk, followed by
paddy straw and sunflower head. The lowest yield (89 g/bag) and
bio-efficiency (13.58%) of P. florida was observed on jackfruit rind.

For centuries,
mushrooms have been used as food and medicine in different cultures.
More recently, many bioactive compounds have been isolated from
different types of mushrooms. Among these, immunomodulators have gained
much interest based on the increasing growth of the immunotherapy
sector. Mushroom immunomodulators are classified under four categories
based on their chemical nature as: lectins, terpenoids, proteins, and
polysaccharides. These compounds are produced naturally in mushrooms
cultivated in greenhouses. For effective industrial production,
cultivation is carried out in submerged culture to increase the
bioactive compound yield, decrease the production time, and reduce the
cost of downstream processing. This review provides a comprehensive
overview on mushroom immunomodulators in terms of chemistry, industrial

The cultivation of Agaricus
bisporus on the spent substrate of
Flammulina velutipes
A large amount of spent substrate produced from the
commercial culturing of edible mushrooms is becoming an
environmental pollution issue. Here spent Flammulina
velutipes substrate, supplemented with unfermented cow
dung, gypsum, and calcium superphosphate, as the main
compost constituent for the cultivation of Agaricus
bisporus were tested. Compared to the conventional
culturing method using rice straw as the primary compost
constituent, the reuse of spent F. velutipes
substrate reduces the cost, shortens the production cycle,
and increases the economic yield of the A. bisporus
culture. Therefore, such recycling is not only practical,
but less environmentally damaging as well.

An improved rapid composting procedure enhance the substrate
quality and yield of Agaricus bisporus
The effect of passive aeration on substrate quality,
composting period and yield was studied using different perforated high-density
polyethylene (HDPE) pipes arrangements, as a composting method for substrate
preparation to Agaricus bisporus cultivation. The ingredients are, mixture of
wheat straw (60%), wheat bran (5.5%), chicken manure (31%), urea (0.5%) and gypsum
(3%) with initial moisture (75%) used for composting. It was demonstrated that
passive aeration is the key parameter controlling composting process, shortened
the compost period, enhance the substrate quality and Agaricus yield as of pile
composting. The substrate physico-chemical characteristics that is, moisture
content, temperature pattern, bulk density, pH, electric conductivity, C: N
ratio and thermophilic fungi activity varied significantly with composting
period. The parallel arrangement (10% perforations) was found best among all
the passive aeration treatments for achieving the quality substrate in shortest
composting period of 16 day and enhance the A. bisporus yield up to 27.6%. This
study also opens possibility to cultivate A. bisporus on a lingo-cellulosic,
non-pasteurized, non-conditioned, aerated substrate and that composting for 16
days improve the mushroom yield with minimum energy, labor and infrastructure.

Abstract
Studies were conducted on the growth and cultivation of Pleurotus pulmonarius
(Fries) singer on lingocellulosic waste. Long composted substrate
formulations of sawdust (86%) + rice bran (10%) + cassava peel (4%)
supported the longest mycelia growth and density. Long composted also
produced larger fruitbodies and significant increase in number of
fruitbodies and compost % yield compared with uncomposted substrate
having the same formulations. The mineral elements of Mg, Cu, Fe, Mn,
Na, K, and P were found to increase from very young to mature fruitbody
of both composted and uncomposted substrate with preponderance of
potassium. Similar results were obtained for crude protein. However,
organic carbon and organic matter tend to decrease from very young to
mature fruitbody of composted and uncomposted substrates.Keywords: Composting, Fructification, Mineral Elements Pleurotus pulmonarius, Organic carbon and Organic matter.

Complete Mitochondrial Genome of the Medicinal Mushroom Ganoderma lucidumAbstractGanoderma lucidum
is one of the well-known medicinal basidiomycetes worldwide. The
mitochondrion, referred to as the second genome, is an organelle found
in most eukaryotic cells and participates in critical cellular
functions. Elucidating the structure and function of this genome is
important to understand completely the genetic contents of G. lucidum. In this study, we assembled the mitochondrial genome of G. lucidum
and analyzed the differential expressions of its encoded genes across
three developmental stages. The mitochondrial genome is a typical
circular DNA molecule of 60,630 bp with a GC content of 26.67%. Genome
annotation identified genes that encode 15 conserved proteins, 27 tRNAs,
small and large rRNAs, four homing endonucleases, and two hypothetical
proteins. Except for genes encoding trnW and two hypothetical
proteins, all genes were located on the positive strand. For the repeat
structure analysis, eight forward, two inverted, and three tandem
repeats were detected. A pair of fragments with a total length around
5.5 kb was found in both the nuclear and mitochondrial genomes, which
suggests the possible transfer of DNA sequences between two genomes.
RNA-Seq data for samples derived from three stages, namely, mycelia,
primordia, and fruiting bodies, were mapped to the mitochondrial genome
and qualified. The protein-coding genes were expressed higher in mycelia
or primordial stages compared with those in the fruiting bodies. The
rRNA abundances were significantly higher in all three stages. Two
regions were transcribed but did not contain any identified protein or
tRNA genes. Furthermore, three RNA-editing sites were detected. Genome
synteny analysis showed that significant genome rearrangements occurred
in the mitochondrial genomes. This study provides valuable information
on the gene contents of the mitochondrial genome and their differential
expressions at various developmental stages of G. lucidum. The results contribute to the understanding of the functions and evolution of fungal mitochondrial DNA.

Chemical, enzymatic and cellular antioxidant activity studies of Agaricus blazei Murrill ABSTRACT Mushrooms
possess nutritional and medicinal properties that have long been used
for human health preservation and that have been considered by
researchers as possible sources of free radical scavengers. In this
work, the antioxidant properties of water extracts from Agaricus blazei Murill, produced by maceration and decoction, are demonstrated in vitro.
Resistance to oxidation is demonstrated through three mechanisms: i)
inhibition of enzymatic oxidative process, with 100% inhibition of HRP
(horseradish peroxidase) and MPO (myeloperoxidase); ii) inhibition of
cellular oxidative stress, with 80% inhibition of the oxidative burst of
polymorphonuclear neutrophils (PMNs); and iii) direct action over
reactive species, with 62% and 87% suppression of HOCl and superoxide
anion radical (O2 ), respectively. From the data, it was concluded that the aqueous extract of A. blazei has significant antioxidant activity, indicating its possible application for nutraceutical and medicinal purposes. Keywords:

ABSTRACT The fruiting bodies of the edible mushroom Lactarius salmonicolor
(Russulaceae) collected from the wild were extracted with solvents of
increasing polarity and evaluated for their antioxidant capacity by
1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging method.
Additionally, their total phenolic content was evaluated by a
modification of the Folin-Ciocalteu method. The methanol extract showed
the most potent radical scavenging activity (inhibition of 36.7% of DPPH
at 3 mg/mL and 6.8 mg gallic acid equivalents/g of dry extract), and
was further fractionated. Total phenolics and antioxidant activity found
to be significant higher in the fractions of increased polarity. The
phytochemical investigation which was performed by chromatographic and
spectrometric techniques led to the isolation of nine compounds from the
methanol extract and the identification of 10 fatty acids and fatty
acids esters from the dichloromethane extract. Evaluation of the
antioxidant activity of isolated compounds showed p-hydroxybenzoic acid (4) to be the most active. Among the various compounds found, the rare azulene type 7-acetyl-4-methylazulene-1-carboxylic acid (1) has been identified, which had been isolated only once before from L. deliciosus. Therefore, this compound seems to have some chemotaxonomic value for Lactrarius species exuding orange to red milk.

مقاله 2013: تاثیر بقایای قارچی استفاده شده در خاک شسسه روی رشد گوجه فرنگی Effect of Mushroom Residue Used in Washed Soil on the Growth of TomatoesIn order to reveal the regulating capacity of mushroom residue on washed
soil, pots experiments were carried out. The growth of tomatoes planted
on washed soil amended by mushroom residue were measured. Mushroom
residue can be helpful to improve the plant height, stem diameter, the
aerial parts fresh weight, root fresh weight, leaf photosynthetic rates
and photosynthesis, and lay a good foundation for the growth of
tomatoes. The effect of mushroom residue is the most significant. Among
all the treatments of adding 2.5%, 5%, 10% mushroom residue, adding 10%
mushroom residue is the best. It can significantly enhance the growth
and photosynthesis of tomatoes, and is among the best of these three
soil treatments for washed soil.

is one of the most common postharvest changes in button mushrooms,
which often results in economic losses. Phenolic compounds, which are
associated with browning, were extracted from the nonbruised and bruised
skin tissue of various button mushrooms with a sulfite-containing
solution and analyzed with UHPLC-PDA-MS. In total, 34 phenolic compounds
were detected. Only small differences in the total phenolic content
between bruising-tolerant and -sensitive strains were observed. The
contents of γ-l-glutaminyl-4-hydroxybenzene (GHB) and γ-l-glutaminyl-3,4-dihydroxybenzene (GDHB) correlated with bruising sensitivity; for example, R2
values of 0.85 and 0.98 were found for nonbruised brown strains,
respectively. In nonbruised skin tissue of the strains with brown caps,
the GHB and GDHB contents in sensitive strains were on average 20 and 15
times higher, respectively, than in tolerant strains. GHB and GDHB
likely participate in the formation of brown GHB–melanin, which seemed
to be the predominant pathway in bruising-related discoloration of
button mushrooms.

The Brazilian almond
mushroom is currently cultivated for its medicinal properties but
cultivars are suspected all to have a common origin. The objective of
this work was to assess the potential of wild isolates of Agaricus subrufescens Peck (Agaricus blazei, Agaricus brasiliensis)
as a source of new traits to improve the mushroom yield and quality for
developing new cultures under European growing conditions. The wild
European strains analysed showed a good ability to be commercially
cultivated on wheat straw and horse manure based compost: shorter time
to fruiting, higher yield, similar antioxidant activities when compared
to cultivars. They have a valuable potential of genetic and phenotypic
diversity and proved to be interfertile with the original culture of the
Brazilian almond mushroom. Intercontinental hybrids could be obtained
and combine properties from both Brazilian and European germplasm for
increasing the choice of strains cultivated by the mushroom growers.
