Plant genetics: The Lingzhi mushroom genome

A team of international researchers led by Shilin Chen and Chao Sun at the Chinese Academy of Medical Sciences and Peking Union Medical College1 have now sequenced the genome of Lingzhi mushroom using the latest sequencing technology and optical mapping approaches. They found that the Lingzhi mushroom genome contains a large assortment of genes and gene clusters, many of which are involved in the production of secondary metabolites.
In particular, the researchers found 78 genes that have important roles in the biosynthesis of triterpenoids — a highly diverse group of steroids with therapeutic properties. By studying the Lingzhi genome, they hope to uncover alternative methods allowing the manufacture of triterpenoids on a large scale.

Abstract
Controlling hyperglycemia and dyslipidemia is associated with reduced risks for diabetic complications. The present study examined the effect of dongchunghacho (Cordyceps militaris; CM) on hyperglycemia and dyslipidemia in an animal model of type 2 diabetes. Fiveweek-old db/db mice were fed an AIN-93G diet or a diet containing 1% CM water extract, whereas db/+ mice were offered the AIN-93G diet for 6 weeks. Consumption of CM significantly decreased blood glycated hemoglobin and serum glucose levels, as well as homeostasis model assessment for insulin resistance (HOMA-IR), in db/db mice. CM significantly lowered serum triglyceride and total cholesterol levels, and increased HDL-cholesterol level. CM water extract inhibited rat intestinal α-glucosidase, with an IC50 of 182 μg/mL in vitro. These results indicate that CM exerted hypoglycemic and hypolipidemic effects in db/db mice.

Agaricus bisporus fucogalactan: Structural characterization and pharmacological approaches
The fucogalactan from A. bisporus (EFP-Ab) obtained on aqueous extraction followed by purification had Mw 37.1x104 g.mol−1 relative to a (1→6)-linked α-D-Galp main-chain partially methylated at HO-3, and partially substituted at O-2 by nonreducing end-units of α-L-Fucp or β-D-Galp. EFP-Ab also inhibited significantly the neurogenic and inflammatory phases of formalin-induced licking, however, the antinociceptive effect was more pronounced against the inflammatory phase with ID50 of 36.0 (25.8–50.3) mg.kg−1. In addition, EFP-Ab decreased the lethality induced by CLP. Its administration reduced the late mortality rate by 40%, prevented neutrophil accumulation in lungs and markedly decreased iNOS and COX-2 protein expression by ileum cells. These data show for the first time that EFP-Ab has significant anti-sepsis, antinociceptive and anti-inflammatory actions, which seems to be related to the decreased iNOS and COX-2 expression. Collectively, the present results demonstrate that EFP-Ab could constitute an attractive molecule of interest for the development of new drugs.

Modulation of Lipoprotein Cholesterol Levels in Plasmodium berghei Malarial Infection by Crude Aqueous Extract of Ganoderma lucidum
In this study, attempt is made to establish changes in serum and liver lipoprotein cholesterols accompanying Plasmodium berghei malarial infection in mice treated with aqueous extract of Ganoderma lucidum at 100, 250, and 500 mg/kg body weight in comparison with 15 mg/kg chloroquine (CQ). Significant increases in all the lipoprotein fractions were observed in infected untreated mice compared with normal control mice. Treatment with 100 and 250 mg/kg G. lucidum extract produced significant reduction in serum total cholesterol (TC) and low-density cholesterol (LDL-C) contents compared with 500 mg/kg G. lucidum and CQ. Treatment with CQ, however, produced significant reduction in hepatic TC and LDL-C compared with the extract. A dose-dependent significant increase in serum high-density lipoprotein cholesterol (HDL-C) was observed in the G. lucidum treated mice compared with normal control but significantly lower compared with CQ-treated mice. Liver HDL-C level was significantly higher in CQ-treated mice compared with normal control and significantly lower compared with G. lucidum-treated and infected untreated mice. A dose-dependent effect of the extract was observed in both serum and liver very-low density lipoprotein cholesterol (VLDL-C). The implication of these results is discussed with respect to the parasite survival and proliferation in the serum and liver.
